Adam (Joseph Gordon-Levitt) is een 27-jarige schrijver van radioprogramma's. Hij wordt gediagnosticeerd met een zeldzame vorm van kanker in zijn wervelkolom. Gezien zijn jonge leeftijd en gezonde levensstijl kan Adam moeilijk geloven dat dit met hem gebeurt. Zonder andere keus besluit hij te beginnen aan chemotherapie en te vechten voor zijn leven. Met de hulp van zijn beste vriend Kyle (Seth Rogen), zijn moeder en de jonge therapeute Katie (Anna Kendrick) van het kankercentrum, leert hij wat en wie de belangrijkste dingen in zijn leven zijn.
